Host cell proteins (HCPs) must be sufficiently removed during downstream processing to ensure product quality and safety. Monitoring is typically performed using ELISA with polyclonal antibodies, whose broad coverage is critical.
 
Here, a quantitative proteomics approach (qIAC-MS) is presented to assess this coverage. Cell culture fluids and process samples were analyzed to identify relevant HCPs.
 
The method complements traditional gel-based techniques and reveals which HCPs are detected or missed by the ELISA, enabling improved risk assessment.
